Epstein-Barr virus (EBV), also known as human herpesvirus 4, is a member of the herpes virus family. The EBV infection prevalence in children under 4 years of age is as high as over 90%. Children infected with EBV mainly present with non-neoplastic conditions, including infectious mononucleosis (IM), chronic active EBV infection (CAEBV), EBV associated hemophagocytic lymphohistiocytosis (EBV-HLH), and X-linked lymphoproliferative syndrome (XLP). EBV is classified as a group 1 carcinogen by the International Agency for Research on Cancer. EBV causes 1% of all cancers worldwide, and EBV-associated cancers account for 5.6% of all infectious cancers. In addition to nasopharyngeal carcinoma, EBV is also associated with a variety of human neoplastic diseases, such as Burkitt's lymphoma (BL), Hodgkin's lymphoma (HL), natural killer (NK)/T cell lymphoma, nasopharyngeal carcinoma (NPC), and some types of gastric cancer.

EBV is mainly transmitted through saliva, but it can also be spread through blood transfusions and organ transplantations. The current laboratory detection methods include serological and nucleic acid testing. Laboratory operators should be trained and qualified for gene amplification or molecular biological testing. The laboratory should be equipped with proper biosafety protection facilities, and proper protection procedures should be established.
